Мікробні індоли на межі взаємодії кишечник-імунна система-мозок за умов стресу: метаболізм, сигнальні шляхи та нейроімунні ефекти

Мікробний метаболізм триптофану, що надходить із їжею, є однією з ключових біохімічних ланок, які поєднують кишкову екосистему, імунітет слизових оболонок, цілісність бар’єрів і функціонування центральної нервової системи за умов стресу. У цьому огляді узагальнено сучасні дані про мікробні індольні метаболіти у межах осі кишечник-імунна система-мозок із чітким розмежуванням прямих причинно-наслідкових доказів, асоціативних спостережень та результатів, отриманих у моделях захворювань, не пов’язаних безпосередньо зі стресом. Утворення індольних метаболітів значною мірою визначається доступністю субстратів, харчовими волокнами, придатними до мікробної ферментації, обміном метаболітами між різними видами мікроорганізмів та pH кишкового вмісту, тоді як сам таксономічний склад мікробіоти не дає повного уявлення про її метаболічну активність. Після утворення індольні похідні взаємодіють із різними рецепторними системами хазяїна – насамперед з арил-вуглеводневим рецептором, а також із рецептором прегнану X та рецептором, активованим проліфераторами пероксисом β/δ. Біологічні ефекти цих сполук істотно залежать від конкретного метаболіту, його концентрації, тканини-мішені та запального контексту, тому мікробні індоли не можна розглядати як універсально захисні постбіотики. Наявні дані свідчать про їхній вплив на симпатоадреналову відповідь, тоді як прямі докази участі окремих індольних метаболітів у регуляції класичної гіпоталамо-гіпофізарно-надниркової осі поки що обмежені. Узагальнення наявних результатів і виявлених методологічних прогалин дає підстави розглядати мікробні індоли як динамічні й контекст-залежні сигнальні молекули, залучені до відповіді організму на стрес, а не як апріорі захисні чинники.
